Nkululeko Habedi, who performed under the name Flabba, died at his family home in Alexandra on Monday morning.
Police told the broadcaster eNCA that the 38-year-old's girlfriend is being questioned on suspicion of murder.
Spokeswoman Katlego Mogale said: "A female suspect in her early 30s has been arrested. The incident was reported in the early hours of the morning."
Flabba was a member of the pioneering hip-hop group Skwatta Kamp which formed in 1996.
In 2007, Flabba won best rap album for his solo project Nkuli Vs Flabba at the annual South African Music Awards.
